abstract Method of determination of nucleotides in food by liquid chromatography of ionic pairs with detection by photodiodes coupled to mass spectrometry of flight time with dual ionization operating in negative mode (HPLC / ESI-APCI-TOF-MS). The method allows the rapid determination of the five nucleotide monophosphates authorized in baby foods (citidine 5? -Monophosphate, uridine 5? -Monophosphate, adenosine 5? -Monophosphate, inosine 5? -Monophosphate and guanosine 5? -Monophosphate), and comprises prior precipitation of food proteins and direct analysis of nucleotides in the resulting extract by HPLC of ionic pairs using isocratic elution. The recovery obtained for fortified samples was satisfactory for all analytes. The proposed procedure allows the determination of the only authorized nucleotides and has been successfully applied to the analysis of different samples of children's and functional foods, such as purees, cereals and pots.
