abstract | An IgG antibody that has a binding affinity for the human CD3 antigen complex and that has a constant region of the human IgG aglycosylated heavy chain and a constant region of the human lambda type chain in whose antibody the heavy chain has a framework of variable region together with CDRs that have the amino acid sequences of SEQ ID NO. 2, 4 and 6 or the respective conservatively modified variants thereof and the light chain has a variable region frame together with CDRs that have the amino acid sequences of the SEQ ID No. 8, 10 and 12 or the respective conservatively modified variants thereof characterized in that the frame of the variable region of the heavy chain corresponds in sequence to the sequence of human type and the frame of the variable region of the light chain corresponds to sequence even frame of the variable region of the rat light chain or derives from a rat frame in which a or more amino acid substitutions selected from the group consisting of Gln-1 to Asp-1; Ala-2 to Phe-2: Val-3 to Met-3; Val-4 to Leu-4; Ala-7 to Pro-7; Asn-8 to His-8; Thr-12 to Glu-12; Leu-14 to Pro-14; Ser-16 to Lys-16; Lys-19 to Ile-19; Leu-20 to Ile-20; Leu-39 to Gln-39; Tyr-40 to Arg-40; Glu-41 to Pro-41; Ser-44 to Ala-44; Met-48 to Val-48; Tyr-50 to Phe-50; Phe-75 to Ser-75, His-79 to Ser-79; Asn-80 to Gly-80; Val-81a Leu-81; Ala-82 to Gln-82; Ile-83 to Thr-83; Ile-88 to Asp-88 and Phe-90 to Tyr-90 whereby the variable region of the light chain includes enough specific amino acids for the rat-like sequence so that the light and heavy chains are associated more strongly than when the region Light chain variable is of the corresponding fully human type. |
