abstract Method for determining the methylation status of at least one cytosine, a methylation pattern, or both in the DNA of a blood sample, a plasma sample, a serum sample or a urine sample of an individual, comprising : (a) providing said sample comprising DNA; (b) isolate the DNA from said sample; (c) treating the isolated DNA with a bisulfite reagent in the presence of a radical scavenger; and (d) determining the methylation status of at least one cytosine in the DNA of said sample, each cytosine being located in a defined position and / or of a methylation pattern in the DNA of said sample, with an amplification reaction based on polymerase and / or an amplification based assay. wherein said bisulfite treated DNA from step (c) is subjected directly to step (d) without prior desulfonation and wherein desulfonation occurs during the initial temperature increase of the amplification reaction.
