Method of quantification of a substrate, comprising the following steps: preparing an analytical element (1) comprising a cavity (4) to accommodate a sample, a working electrode (2) and a counter electrode (3) exposed towards the inside of said cavity, a reactive layer comprising at least one oxidoreductase and which is formed inside or in the vicinity of said cavity, a sample port (9) from which the sample is introduced into the cavity, a ventilation duct (5) open in the vicinity of the cavity and a puncture element (6); puncturing the subject's skin through the puncture element, while both the sample feed port and the ventilation duct are closed by the puncture element; move the puncture element into a position, in which the ventilation duct is open to cause a body fluid to be introduced into the cavity; and move the puncture element to a position, in which the puncture element closes the ventilation duct and the sample feed port and its pointed end does not protrude from the feed port to the outside of the analytical element, apply a voltage between said working electrode and said counter electrode; and detect a current flowing through the working electrode to measure the concentration of the substrate contained in the body fluid. |
