abstract |
A method for manufacturing an immunogenic conjugate comprising a Streptococcus pneumoniae serotype 3 polysaccharide covalently linked to a transporter protein, the method comprising: (a) reacting a purified serotype 3 polysaccharide with a weak acid resulting in a serotype polysaccharide 3 hydrolyzate; (b) reacting the hydrolyzed serotype 3 polysaccharide with an oxidizing agent in the presence of bivalent cations resulting in an activated serotype 3 polysaccharide in which the oxidizing agent is periodic acid and the bivalent cations are Mg2 + or Ca2 +; (c) combining the activated serotype 3 polysaccharide with a carrier protein; (d) reacting the activated serotype 3 polysaccharide and the transporter protein combined with a reducing agent, resulting in a serotype 3 polysaccharide conjugate: transporter protein; and (e) protecting aldehydes that have not reacted in the serotype 3 polysaccharide conjugate: transporter protein resulting in an immunogenic conjugate comprising the serotype 3 polysaccharide of Streptococcus pneumoniae covalently linked to a transporter protein. |
