A method of obtaining a desired gene product comprising culturing a host cell, wherein the host cell comprises a vector, and the vector comprises a polynucleotide, said polynucleotide comprising: (i) an extended methylation-free CpG island comprising promoters dual or bi-directional that are transcribed divergently, in which the CpG island free of extended methylation extends over more than 300 bp and also comprises an average GC content of 60%; (ii) an expressible gene encoding a desired gene product, in which the expressible gene is operably linked to the CpG island; and (iii) a promoter, operably linked to the expressible gene, in which the promoter is not operably linked in nature to the CpG island; in which the CpG island free of extended methylation opens the chromatin or keeps the chromatin in an open state and facilitates the activation of the reproducible transcription of the expressible gene. |
