abstract | Method of modifying a therapeutic polypeptide, comprising: (a) identifying at least one immunodominant epitope in the polypeptide using an antibody or population of antibodies obtained from an untreated human or untreated animal or a population thereof, where the Polypeptide has an amino acid sequence identical to the native sequence of an endogenous polypeptide homologue in the same human or the same animal species or a biologically active fragment of the endogenous polypeptide, where the antibody or antibodies do not substantially inhibit a therapeutic activity of the polypeptide; and the immunodominant epitope is not located in a region of the polypeptide that provides a therapeutic activity of the polypeptide; and (b) modify the immunodominant epitope to reduce an immune response to the polypeptide while maintaining a substantial therapeutic activity of the polypeptide. |
