abstract | Procedure for the isolation and separation of single and double stranded nucleic acids from a sample containing these nucleic acids, wherein the double stranded nucleic acid is DNA and the single stranded nucleic acid is RNA, and the nucleic acids of Single and double strands are linked to a support material from which double stranded nucleic acids are eluted with a first elution solution and single stranded nucleic acids are then eluted with a second elution solution, characterized in that the elution of double stranded nucleic acids are carried out in the presence of at least one divalent metal ion in a concentration at least so high that during the elution of double stranded nucleic acids no single stranded nucleic acids are eluted, wherein the first elution solution is a buffer with low salt or water content, and the second elution solution is a solution n with low salt content or water and wherein the at least one divalent metal ion selected from the group of alkaline earth metals, and their concentration is not greater than 200 mM. |
