abstract | A method for purifying AAV vector particles from an AAV preparation comprising AAV vector particles and empty AAV capsids, to provide a product substantially free of empty AAV capsids, said method comprising: a) providing a host cell comprising AAV vector particles; b) lyse said host cell to obtain a crude cell lysate comprising AAV vector particles and empty AAV capsids; c) applying said crude cell lysate to a first cation exchange chromatography column under conditions in which said AAV vector particles and said empty AAV capsids are attached to the column; d) eluting said AAV vector particles and said AAV empty capsids under non-separation conditions to provide an AAV preparation comprising AAV vector particles and empty AAV capsids; e) applying the AAV preparation from (d) to a second column of cation exchange chromatography under conditions under which said AAV vector particles and said empty AAV capsids are attached to the column; f) adding a low salt buffer to the column of (e) under conditions in which the AAV vector particles are eluted and the empty AAV capsids remain attached to the column; and g) collecting the eluted fractions of (f) comprising AAV vector particles to provide an AAV product substantially free of empty AAV capsids. |
