Method for enzymatically treating nucleic acid molecules in a mixture that includes a chaotropic agent, degraded and denatured proteins and DNA and RNA molecules released from bound proteins, comprising diluting said mixture with at least one aqueous reagent to reduce the concentration. of the chaotropic agent less than 0.05 M, preferably less than 0.01 M, and subject at least some of said RNA and DNA molecules to at least one enzymatic treatment without extracting or isolating said RNA and DNA molecules from each other, or from the other components of the reaction mixture. |
