abstract | An in vitro method of site-specific integration of a sequence of polynucleotides of interest in a genome of a cell of a multicellular organism, said method comprising: introducing (i) a circular director construction comprising a first recombination site and the polynucleotide sequence of interest, and (ii) an altered specific unidirectional site recombinase in the cell, in which the genome of said cell comprises a second natural recombination site for the genome and recombination between the first and second recombination site is facilitated by the recombinase altered, wherein said first recombination site is an attB recombination site and said second recombination site is an attP recombination pseudosite, keeping the cell in conditions that allow recombination between said first and second recombination site, the recombination being mediated by altered recombinase and always or the result of recombination the site-specific integration of the polynucleotide sequence of interest in the cell genome, in which the unidirectional site-specific altered recombinase is constituted by a polypeptide sequence that is at least 80% identical to the natural recombinase C31 of SEQ ID NO: 21 (other than SEQ ID NO: 21 itself) and has integrase activity, and in which the unidirectional site-specific altered recombinase has a specificity of DNA binding or an activity level that differs from that of a natural unidirectional site-specific recombinase and may mediate recombination between an attP recombination pseudosite and an attB recombination site. |
