abstract | A method for generating a human tear lipocalin mutein, in which the mutein binds to a given unnatural ligand of human tear lipocalin with detectable binding affinity, comprising: (a) subjecting a nucleic acid molecule that encodes a human tear lipocalin to mutagenesis in at least 12, 14 or 16 of the codons of any of the positions of amino acid sequence 26-34, 56-58, 80, 83, 104-106 and 108 of the linear polypeptide sequence of the native mature human tear lipocalin, in which at least one of the codons encoding cysteine residues at sequence positions 61 and 153 of the linear polypeptide sequence of the mature human tear lipocalin has been mutated to encode any other amino acid residue , thereby obtaining a plurality of nucleic acids encoding human lacrimal lipocalin muteins, (b) expressing the one or more mutein nucleic acid molecules obtained s in (a) in an expression system, thereby obtaining one or more muteins and (c) enriching the one or more muteins obtained in step (b) and having binding affinity detectable by a given unnatural ligand of human tear lipocalin by means of selection and / or isolation, provided that the unnatural ligand of human tear lipocalin is not the CD4 human T lymphocyte co-receptor. |
