abstract Plant of the Allium cepa species that is resistant to downy onion powdery mildew caused by the fungus Peronospora destructor (Pd) due to a locus of resistance to Pd present homozygously on the long arm of chromosome 3 in the genome of said plant, in in which the locus of resistance to Pd is the introgression fragment of Allium roylei in the genome of a plant of line 3591-1, registration number NCIMB 41249, and in which the following procedure is carried out: a) restricting the genomic DNA of said plant with the restriction enzymes PstI and MseI; b) perform a ligation with the following oligonucleotide adapters: alpha 5'-CTCGTAGACTGCGTACATGCA CATCTGACGCATGT-5 ', and beta 5'-GACGATGAGTCCTGAG TACTCAGGACTCAT-5'; c) selectively amplify sets of restriction fragments, characterizing the presence of the Pd resistance locus by &8226; a 61 nucleotide amplification product when the following primers are used in step c): (A) 5'-GACTGCGTACATGCAGAAC and 5'-GATGAGTCCTGAGTAACTT, &8226; a 151 nucleotide amplification product when the following primers are used in step c): (B) 5'-GACTGCGTACATGCAGAAG and 5'-GATGAGTCCTGAGTAAAAC and &8226; a 330 nucleotide amplification product when the following primers are used in step c): (C) 5'-GACTGCGTACATGCAGACA and 5'-GATGAGTCCTGAGTAACCA.
