abstract Method for the identification of a pathogenic organism from a predetermined group of pathogens, comprising: a) at least partially purifying a nucleic acid from a clinical sample, b) submitting at least a first aliquot of said clinical specimen to at least one amplification and detection reaction in a reaction vessel, comprising: ba) an amplification step using a first set of amplification primers capable of amplifying a preselected spacer region 16s / 23s or 18s / 26s of several or all members of said predetermined group of pathogens, bb) a detection step using at least 2, 3 or multiple hybridization reagents, said reagents being jointly capable of specifically detecting a preselected spacer region 16s / 23s or 18s / 26s of among all the members of said group of pathogens, said detection stage comprising bb) the stages: bb a) monitoring the hybridization of each of said hybridization reagents at a preselected temperature, said hybridization being indicative of at least the genus of said pathogen present in the sample, and bbb) monitoring the temperature dependence of the hybridization with said hybridization reagents, and determination of the melting temperature, said temperature dependence being indicative of at least the species of said pathogen. c) Determine if a hybridization signal is produced in stage bba), if a pathogenic organism of a particular genus is present in said sample, and determine from which temperature dependence of stage bbb), which pathogenic species of said genus is present in said sample.
