Sequencing method of a single target nucleic acid molecule having a plurality of nucleotide bases comprising: providing a complex of an enzyme that polymerizes the nucleic acid and the target nucleic acid molecule to allow the formation of a nucleic acid chain in complementary growth to the target nucleic acid; providing a plurality of types of labeled nucleotide analogs, in which each type of nucleotide analog is complementary to a different nucleotide in the target nucleic acid sequence; polymerize the plurality of types of labeled nucleotide analogs incorporating the plurality of types of labeled nucleotide analogs in the growing nucleic acid chain, in which each of the incorporated analogs can be polymerized with a subsequent nucleotide analog to allow continuous polymerization of a plurality of nucleotide analogs; identify the time sequence of incorporation of said plurality of types of labeled nucleotide analogs in the growing nucleic acid chain by detecting the residence times of the labeled nucleotide analogs close to the enzyme that polymerizes the nucleic acid in real time while the template-directed primer extension takes place, and the marker being removed after detection. |
