abstract | Process for preparing an insulin compound where a) in a reactive mixture containing at least about 55%, preferably at least about 60%, more preferred at least 70%, of water (weight / weight), an insulin precursor is subject to an enzymatic cleavage and, after that, b) the intermediate product is coupled with a nucleophilic compound in the reaction mixture used for the enzymatic cleavage reaction with the proviso that the composition of the reactive mixture has been modified so that the water content in the reaction mixture is in the range of about 10% to about 50% water (weight / weight), preferably in the range of about 20% to about 40% water (weight / weight), and c), if desired, eliminating the protection group (s), and where no isolation of the intermediate product is performed between the cleavage phase (a) and the coupling phase (b). |
