abstract A high performance method for determining the activity of a phospholipase enzyme associated with lipoprotein A2 (Lp-PLA2) in a set of samples comprising the steps of; (i) prepare a solution comprising labeled platelet activation factor (PAF); (ii) take aliquots of each of the samples in the set into a well of a microtiter plate or microcentrifuge tube; (iii) contacting each of the tissue samples in the assembly with the solution of the preparation stage; (iv) contacting each of the solutions of the first stage of contact with a PAF sequestrant molecule to form a PAF sequestrant molecular complex; (v) contacting said PAF sequestrant molecular complex with a precipitation solution comprising trichloroacetic acid (TCA) to form a precipitate and a supernatant; (vi) removing said PAF sequestrant molecular complex by centrifuging said microtiter plate or microcentrifuge tube at at least 6,000 g for at least 5 minutes at less than 10 ° C; and (vii) detect Lp-PLA2 activity, where at least one sample has been taken from an animal that has been given an Lp-PLA2 inhibitor.
