abstract | A method of preparing a compound of Formula (I): ** (See formula) ** or a pharmaceutically acceptable enantiomer, diastereomer, hydrate, solvate or salt thereof, consisting of the reaction of a compound of formula (IV): ** (See formula) ** with an acid under reducing conditions, in a non-basic organic solvent, at a temperature between approximately room temperature and the reflux temperature of the solvent, to obtain the compound of Formula (I), where Ar is: A) phenyl optionally mono-, di- or tri-substituted with Ra or disubstituted in adjacent carbons with -O-C 1-4 alkylene -O-; each Ra is independently selected from the group consisting of -OH, -C1-6 alkyl, -O-C1-6 alkyl, -CN, -NO2, -N (Rb) Rc, -C (O) N (Rb) Rc , -N (Rd) C (O) Rd, -N (Rd) SO2-C1-6 alkyl, -C (O) C1-6 alkyl, -S (O) 0-2-C1-6 alkyl, -SO2N (Rb) Rc, -SCF3, halo, -CF3, -OCF3, -COOH, -COO-C 1-6 alkyl and tetrazolyl, where Rb and Rc are each independently -H or -C 1-6 alkyl; and where Rd is -H or -C 1-6 alkyl; B) a monocyclic aromatic hydrocarbon group having five ring atoms, having a carbon atom that is the point of attachment, having a carbon atom substituted by> O,> S,> NH or> N (C1- alkyl) 4), which has up to an additional carbon atom eventually substituted by -N = and possibly mono- or di-substituted with Ra; or C) a monocyclic aromatic hydrocarbon group having six ring atoms, having a carbon atom that is the point of attachment, having one or two carbon atoms substituted by -N = and possibly mono- or di-substituted with Ra; ALK is: i) C1-7 alkyl, optionally substituted with Re, where Re is selected from the group consisting of -OH, -O-C1-6 alkyl, -CN, -N (Rf) Rg, -C (O) N (Rf) Rg, -N (Rh) C (O) Rh, -N (Rh) SO2-C1-6 alkyl, -C (O) -C 1-6 alkyl, -S (O) 0-2-alkyl C1-6, -SO2N (Rf) Rg, fluoro, -CF3, -COOH and -COO-C1-6 alkyl, where Rf and Rg are each independently -H or -C1-6 alkyl and where Rh is -H or -C 1-6 alkyl; ii) a cycloalkyl ring, optionally mono-, di- or tri-substituted with Re or C1-6 alkyl; iii) phenyl optionally mono-di- or tri-substituted with Re; or iv) -C1-2-CYC alkyl; where CYC is selected from the group consisting of: a) a phenyl or naphthyl group, optionally mono-, di- or tri-substituted with Rh or disubstituted in adjacent carbons with -O-C 1-4 alkylene; each Rh is independently selected from the group consisting of -OH, -C1-6 alkyl, -O-C1-6 alkyl, -CN, -NO2, -N (Ri) Rj, -C (O) N (Ri) Rj , -N (Ri) C (O) Rj, -N (Ri) SO2-C1-6 alkyl, -C (O) -C1-6 alkyl, -S (O) 0-2-C1-6 alkyl, - SO2N (Ri) Rj, -SCFa, halo, -CF3, -OCF3, -COOH and -COO-C1-6 alkyl, where Ri and Rj are each independently -H or -C1-6 alkyl; and b) a monocyclic aromatic hydrocarbon group having five ring atoms, having a carbon atom that is the point of attachment, having a carbon atom substituted by> O,> S,> NH or> N (C1- alkyl) 4), which has up to an additional carbon atom eventually substituted by -N = and eventually mono- or di-substituted with Rh; c) a mono-cyclic aromatic hydrocarbon group having six ring atoms, having a carbon atom that is the point of attachment, having one or two carbon atoms substituted by -N = and possibly mono- or di-substituted with Rh; and d) a cycloalkyl ring, which has 0, 1 or 2 unsaturated bonds, optionally mono- or di-substituted with Rh; R2 is -CO2R10 or an unsubstituted or substituted benzyl group with one or two substituents -OCH3, where R10 is methyl, ethyl, 2-trimethylsilylethyl, 2-phenylethyl, 1- (1-adamantyl) -1-methylethyl, t-butyl , 1-adamantyl, vinyl, allyl, benzyl, p-methoxybenzyl, p-nitrobenzyl, p-chlorobenzyl, 2,4-dichlorobenzyl or diphenylmethyl; and P1 and P2 are each independently -C1-4 alkyl or, alternatively, P1 and P2 taken together form - (CH2) 2-3-. |
