abstract | Method that uses a protein complement / universal indicator system (PAC / URS) assay for the detection and localization of a ligand of a cellular receptor, including said method: (a) the generation of a first nucleic acid vector encoding a first fusion product, comprising: (i) a first fragment of a first PCA / URS indicator molecule, and (ii) a second molecule, fused with said first fragment, comprising a first subdomain of the erythropoietin receptor; (b) the generation of a second nucleic acid vector encoding a second fusion product, comprising: (i) a second fragment of said first PCA / URS indicator molecule, and (ii) a third molecule, fused with said second fragment, comprising a second subdomain of said erythropoietin receptor, and wherein said second subdomain may be the same as said first subdomain, or different from said first subdomain; (c) transfection of prokaryotic or eukaryotic cells with said first and second nucleic acid vectors; and (d) verification in said transfected cells of the PCA / URS indicator activity, said activity indicating the first and second fragment of the PCA / URS molecule mediated by the interaction of said first and second erythropoietin receptor subdomains, said association being induced by linking said receptor with said ligand, wherein said PCA / URS indicator molecule is a polypeptide encoded by a gene that has been dissected rationally into at least two fragments encoding, respectively, said first fragment and said second fragment of said PCA / URS indicator molecule. |
