titleOfInvention NEW SYNTHESIS PROCEDURE OF (7-METOXI-1-NAFTIL) ACETONITRILE AND ITS APPLICATION TO THE SYNTHESIS OF AGOMELATIN.
abstract Industrial synthesis process of the compound of formula (I) ** (See formula) ** characterized in that 7-methoxy-1-tetralone is reacted, of formula (IV): ** (See formula) ** with cyanoacetic acid , of formula (V): ** (See formula) ** in conditions of elimination of the water formed, in the presence of a catalytic amount of the compound of formula (VI): ** (See formula) ** where R and R ' ', identical or different, each represents a linear or branched (C3-C10) alkyl group, an unsubstituted or substituted aryl group or a linear or branched unsubstituted or substituted aryl (C1-C6) group, to conduct, after filtration and washing with a basic solution, the (7-methoxy-3,4-dihydro-1-naphthalenyl) acetonitrile of formula (VII): ** (See formula) ** compound of formula (VII) which is subjected to reaction with a hydrogenation catalyst in the presence of an allyl derivative, to lead to the compound of formula (I), after filtration and evaporation of the solvent nte, compound of formula (I) which is isolated in the form of a solid after recrystallization, it being understood that: - by aryl is meant a phenyl, naphthyl or biphenyl group; - the term "substituted" attributed to the terms "aryl" and "arylalkyl" means that the aromatic part of these groups may be substituted with 1 to 3 groups, identical or different, selected from linear or branched (C1-C6) alkyl, hydroxyl and linear or branched (C1-C6) alkoxy; - "allyl derivative" means any molecule that contains 3 to 10 carbon atoms and that may contain in addition to 1 to 5 oxygen atoms, and that contains at least one part -CH2-CH = CH2.
