METHOD FOR THE IDENTIFICATION OF POTENTIALLY USEFUL COMPOUNDS IN THE TREATMENT OR PREVENTION OF NEURODEGENERATIVE DISEASES. |
abstract |
Method for the identification of compounds thatninduce or inhibit endoplasmic reticulum stress or stressnoxidativen n n The present invention relates to a methodnfor the identification of producing compounds or inhibitors ofnendoplasmic reticulum stress or oxidative stress. Sayingnmethod is based on the transformation of cells stablynwith a vector that expresses a reporter gene under the control of the genenDDIT3 (also called CHOP). This promoter includes thennucleotides from -1626 to +60 of said gene, having two variantsnof it: -1507 C / G. While the variant-1507nC is not activated under conditions of oxidative stress, -1507 GnRespond to this stimulus in an accused manner. On the other hand, bothnpromoters are activated in reticulum stress situationnendoplasmic, or in a combined situation of both conditions. |
