A method of in situ detection of an immunohistochemical epitope or a nucleic acid sequence of interest in a biological sample, comprising binding a conjugated molecule labeled with an enzyme to the epitope or sequence of interest in the presence of a reducing species without redox activity and a soluble metal ion, wherein said reducing species without redox activity is a substrate for said conjugate molecule labeled with an enzyme and wherein said enzyme of said conjugated molecule labeled with an enzyme catalyzes the transformation of the reducing species without redox activity into a reducing agent, thereby reducing the metal ion to insoluble metal particles in oxidation state 0, at or around the point where the enzyme is anchored. |
