titleOfInvention | CONJUGATES OF MACROLIDES WITH ANTIINFLMATORY ACTIVITY. |
abstract | A compound according to Formula I: ** (See formula) ** in which M represents the macrolide subunit of substructure II: ** (See formula) ** in which R1, R2, R3, R4 and R5 are chosen , independently of each other, from the group consisting of hydrogen, C1-C4 alkyl, alkanoyl, alkoxycarbonyl, arylmethoxycarbonyl, aroyl, arylalkyl, alkylsilyl and alkylsilylalkoxyalkyl; RN represents the covalent bond with X1 of the L chain; L represents the substructure III chain: -X1 - (CH2) m - Q - (CH2) n - X2 III in which X1 is -CH2- or -C (O) -; X2 is -NH- or -O-; Q is -NH- or -CH2-; the symbols m and n, independently, are integers from 0 to 4; with the proviso that if Q = NH, n cannot be 0; D represents a non-steroidal anti-inflammatory subunit or a steroid subunit or a pharmaceutically acceptable salt or solvate thereof; wherein "alkyl", unless otherwise indicated, means a linear or branched saturated monovalent hydrocarbon radical of one to ten carbon atoms, which may be substituted with up to five substituents including halogen, hydroxy, alkoxy, acyl, acylamino, cyano, amino, N-C1-C4 alkyl, amino, N, N-di (C1-C4 alkyl) amino, aryl or teroaryl, thiocarbonylamino, acyloxy, amidino, alkylamidino, thioamidino, aminoacyl, aminocarbonylamino, aminothiocarbonylamino, aminocarbonyloxy, aryl, heteroaryl , aryloxy, aryloxyaryl, nitro, carboxy, carboxy alkyl, alkyl substituted with carboxy, carboxycycloalkyl, cycloalkyl substituted with carboxy, carboxyaryl, aryl substituted with carboxy, carboxyheteroaryl, heteroaryl substituted with carboxy, carboxyheterocyclyl, cycloalkyl, heterocyclyl, cycloalkyl, heterocyclyl, cycloalkyl, heterocyclyl heteroaryloxy, heterocyclyloxy, and oxycarbonylamino; and the present definition of alkyl applies to other groups having an alkyl moiety such as alkoxy or alkanoyl; "cycloalkyl" means a cyclic group having 3-8 carbon atoms having a single ring optionally fused to an aryl or heteroaryl group, and which may be substituted as specified for "aryl" below; "aryl" means an unsaturated aromatic carbocyclic group having 6-14 carbon atoms, having a single ring or several condensed rings and which may be optionally condensed with an aliphatic or aryl group or may be substituted with one or more substituents selected from halogen, hydroxy, C1-C7 alkyl, C1-C7 alkoxy or aryloxy, C1-C7 alkylthio or arylthio, alkylsulfonyl, cyano and primary or non-primary amino; "heteroaryl" means a monocyclic or bicyclic aromatic hydrocarbon ring having 2 to 10 carbon atoms and 1 to 4 heteroatoms, such as O, S or N, and which may optionally be condensed with another heteroaryl, aryl or aliphatic cyclic group , and which may be substituted with the same substituents specified above for aryl; "heterocyclic" means a saturated or unsaturated group that has only one or several rings and 1 to 10 carbon atoms and 1-4 heteroatoms selected from nitrogen, sulfur or oxygen, where the other or the ones in a condensed ring system other rings may be aryl or heteroaryl, and wherein the heterocycle may be substituted as specified for alkyl groups above. |
