A method of producing an active human IL-18 polypeptide from a human IL-18 precursor polypeptide, comprising: (i) Bicistronically co-expressing caspase 4 or caspase 5 with the human IL-18 precursor polypeptide using a bicistronic expression vector comprising a sequence encoding the human IL-18 precursor followed by a sequence encoding caspase 4 or caspase 5 separated by an intergenic region that includes a Shine-Delgarno sequence; and (ii) Purify the active polypeptide. |
