abstract The use of a viral vector to prepare a pharmaceutical composition for the treatment or prophylaxis of a mammal by gene therapy of muscle parenchymal cells inside a limb, characterized in that the gene therapy is effected by obstructing the inflow and outflow of a blood vessel of said member, and inserting said viral vector into said blood vessel, whose insertion comprises injecting said viral vector, selected from the group consisting of adenoviral, adeno-associated viral and retroviral in solution, from 10 seconds to 30 minutes, in said blood vessel thereby increasing the hydrostatic pressure on the wall of said blood vessel, and wherein said obstruction is maintained for a period of time sufficient to allow administration of the viral vector.
