abstract | A phage particle having on its surface a single chain TCR polypeptide (scTCR) or a dimeric TCR polypeptide pair (dTCR), wherein said scTCR is constituted by a first segment a sequence of the variable domain of the alpha chain or delta of the TCR fused with the N-terminus of an extracellular sequence of the constant domain of the alpha chain of the TCR, a second segment consisting of a variable domain sequence of the beta or gamma chain of the TCR fused with the N-terminus of an extracellular sequence of the constant domain of the TCR beta chain, a spacer sequence that links the C-terminus of the first segment with the N-terminus of the second segment, or vice versa, and a disulfide bridge between the first and second segments, wherein said disulfide bridge is one which has no equivalent in native alphabeta or gammadelta T cell receptors; wherein said dTCR polypeptide pair is constituted by a first polypeptide in which a sequence of the variable domain of the alpha or delta chain of the TCR fused with the N-terminus of an extracellular sequence of the constant domain of the alpha chain of the TCR a second polypeptide in which a sequence of the variable domain of the TCR beta or gamma chain is fused with the N-terminus of a sequence corresponding to the extracellular sequence of the constant domain of the TCR beta chain, the first and second polypeptides, in which they are linked by a disulfide bridge that has no equivalent in the native alphabeta or gammadelta T cell receptors, wherein said disulfide bridge between the first and second segments of the scTCR, or between the first and second polypeptides of the dTCR, between the substituted tanks for the following pairs of residues in exon 1 of TRAC * 01 for the TCR alpha chain and TRBC1 * 0 or TRBC2 * 01 for the TCR beta chain: |
