abstract | Process for the production of amidated peptides at the C-terminal of general formula II (AS) n-Xm-NH2 (formula II) in which AS means one or more genetically encodable amino acids; n is from 5 to 2000, preferably from 10 to 1000, in particular from 15 to 500, very especially preferably from 20 to 400; and (AS) n and / or (AS) n-Xm preferably means a biologically active peptide or protein, X means one or more basic amino acids or one of its derivatives, preferably lysine, histidine and / or arginine, in particular lysine; m is from 1 to 15, preferably from 3 to 10, in particular from 6 to 8; ynym means whole numbers and in which a) host cells are grown in a suitable nutrient medium, b) a compound of general formula I is expressed: (AS) n-XmYp (formula I), in which Y means one or more neutral amino acids, preferably glycine, p is 1 to 10, preferably 1 to 5, in particular 1; a) optionally the compound of the formula I is released from a suitable precursor peptide by enzymatic or chemical cleavage; b) the expression products of step b) or the intermediate products of step c), possibly after a purification step, are reacted with an alpha-amidant enzyme (PAM); and c) the peptide amidated at the C-terminal of the general formula II is suitably purified, preferably by a preparative chromatographic method. |
