abstract | Protein domains of affinity to choline fornimprove expression, immobilization and purification ofnpolypeptidesn n n Objects of the present invention are a polypeptide sequence derived from that of the choline-binding module of the LytA amidase of Streptococcus pneumoniae , as well as a set of expression vectors intended for obtaining, from translational fusions to the coding sequences of said polypeptide in Bacteria and yeast. Various structural and purification process improvements are also objects of the invention, which allow to improve the expression and stability of the fusions, in addition to minimizing interference with the fused polypeptide and optimizing the proteolytic separation of said module. As examples of embodiment, the construction of a set of plasmid vectors with the operative signals necessary for the expression in bacteria or in yeasts of one of these sequences is described, and which allow their fusion to heterologous genes to obtain hybrid proteins that can be immobilized in a variety of supports, and eventually purified by affinity chromatography. |
