abstract A procedure for the determination of microbiological populations in an opaque medium, characterized by comprising: (a) obtaining two Aliquots of material from the opaque medium; (b) add a Fluorogenic Dye to the first Aliquot, Aliquot Part referred to as the Aliquot-Dye Part, and add a Metabolic Inhibitor to the second Aliquot, followed by the addition of a Fluorogenic Dye to the second Aliquot Part, second Aliquot Part which is now referred to as Aliquot-Inhibitor-Coloring Part; (c) allowing said Fluorogenic Dye to be reduced by the dehydrogenases present in the membranes of the microbiological organisms present to produce a Reacted Fluorogenic Dye, said Fluorogenic Dye and said Reacted Fluorogenic Dye having different fluorescent signals; (d) providing means for measuring the fluorescent signals in said Aliquot-Dye Part and in said Aliquot-Inhibitor-Dye Part, the fluorescent signals being measured at the emission wavelength of the Fluorogenic Dye and at the emission wavelength of the Reacted Fluorogenic Dye; (e) using said measuring means to measure the fluorescent signals in said Aliquot-Dye Part and in said Aliquot-Inhibitor-Dye Part at the emission wavelength of the Fluorogenic Dye and at the emission wavelength of the Reacted Fluorogenic Dye, while the values of measured fluorescent signals that are below a predetermined noise level are discarded; (f) calculate the ethyl RATIO, the RATIO of the fluorescent signal of the Reacted Fluorogenic Dye being adjusted for the interactions with chemical products, the fluorescent signal for the Fluorogenic Dye adjusted for interactions with the chemicals; and (g) use the ETERNAL RATIO to determine the degree of microbiological contamination in said opaque medium.
