abstract | A method for the preparation of polynucleotide fragments for use in polynucleotide rearrangement, comprising: obtaining a homologous polynucleotide library from a wild type gene by successive controlled mutagenesis steps denaturation and hybridization of said polynucleotide for the formation of the heteroduplex polynucleotides the division of said heteroduplex polynucleotides using any of the polynucleotide repair system proteins that can divide the base pairs with in vitro replication error comprising DAM methylase, MutS, MutL, MutH, exonuclease, DNA helicase II, SSB protein, DNA polymerase III, DNA ligase, or a combination thereof; or with a base division repair complex consisting of glycosylase DNA, AP endonuclease, DNA polymerase I, DNA ligase, or a combination thereof; or with a nucleotide division repair complex consisting of Uvr-A, Uvr-B, Uvr-C, DNA polymerase I, DNA ligase, or a combination thereof denaturing said heteroduplex divided polynucleotides to obtain fragments. |
