abstract A method for manufacturing a nucleic acid molecule comprising the steps of a) providing a first at least partially double-stranded oligonucleotide having a modification that allows the oligonucleotide to engage a surface, in which the oligonucleotide comprises a recognition site of a first IIS type restriction enzyme that cuts out of its recognition site, and said oligonucleotide comprises a single stranded projection, b) providing a second at least partially double stranded oligonucleotide, wherein the oligonucleotide comprises a recognition site or a part of the same or a sequence that is complementary thereto of a second restriction enzyme of type IIS that cuts out of its recognition site, and said second oligonucleotide comprises a single stranded projection, c) ligating the first and second oligonucleotides by its projections, generating a first ligation product, d) immobilize the first surface ligation product by modification, e) cutting the immobilized ligation product with the first IIS type restriction enzyme, thereby releasing an elongated oligonucleotide having a projection, f) combining the elongated oligonucleotide with an additional oligonucleotide at least partially double stranded which has a modification that allows the oligonucleotide to engage a surface, in which the additional oligonucleotide comprises a recognition site of an additional IIS type restriction enzyme that cuts outside its recognition site, and said oligonucleotide comprises a single-chain projection, and linking the second elongated oligonucleotide and the additional oligonucleotide at least partially double-stranded by its projections, forming an additional ligation product, g) immobilizing the additional ligation product on a surface by modification, h) cutting the product from linkage adds l with the additional IIS type restriction enzyme, releasing an elongated oligonucleotide having a protrusion, and i) optionally, repeat steps f) to h).
