abstract |
A DNA molecule that encodes a fusion protein comprising: (a) a cleavage site of the target protease; (b) a ligand binding domain; and (c) a DNA binding domain; wherein the association of a ligand with said fusion protein mediates the binding of the fusion protein to a ligand response element ("ERL") operably linked to an indicator gene; wherein said protease cleavage site of said fusion protein can be cleaved so that none of the resulting fusion protein fragments is capable of modulating the expression of said reporter gene; and wherein the fusion protein also comprises an expression modulator domain or is associated with a second protein that has an expression modulator domain, wherein said expression modulator domain regulates the transcription of said reporter gene; and wherein said cleavage site of the target protease is located in the DNA binding domain or in the modulator domain, if present, or between any of the domains of said fusion protein. |
