abstract | A method for producing a protein characterized in that said protein is a fragment of a protein that is a primary product of the translation of a target gene that is the C or N-terminal end of said primary product of the translation of a target gene and in that said fragment exerts a biological function, the method comprising: (i) growing a host cell transfected with a DNA construct comprising: (a) a regulatory DNA sequence capable of initiating transcription and translation or terminating transcription and the translation of the DNA encoding said fragment; and (b) a region of targeting DNA comprising sequences homologous to a region of the target gene in 5 '' or 3 '', respectively, with respect to the sequence encoding said fragment, the construction being integrated into the genomic DNA of the cell host at a position determined by the targeting DNA segment so that the expression of said fragment is under the control of the regulatory DNA; and, optionally (ii) culturing the recombinant cell in a homologous manner; and, optionally (iii) collecting said fragment; wherein said fragment is selected from the group consisting of: (i) an extracellular binding domain of a membrane receptor; (ii) a proteolytic fragment of an extracellular matrix structural protein that has antiangiogenic properties; and (iii) a proteolytically released protein from an inactive precursor protein. |
