abstract | A method for detecting the presence of a difference between a target nucleic acid sequence and a reference nucleic acid sequence, comprising: (a) amplification of the target nucleic acid sequence by polymerase chain reaction, using a P2 primer, a P4 primer with a 3 '' region capable of hybridizing to the target sequence and a T region in the 5 '' tail that is not complementary to the target sequence, and a P5 primer that is capable of hybridizing to the target sequence at a 3 '' location of the sequence capable of hybridizing to said 3 '' fraction of primer P4; in which either: primer P2 is a mixture of primer P2 with a first marker and primer P2 with a second marker, or primer P4 has a first marker and primer P5 has a second marker; (b) the formation of a target partial duplex with tail of the target sequence, said partial duplex having one of said first marker or said second marker, and a tail of two non-complementary regions in which a first region is the P5 sequence or its complement and the second region is the sequence T or its complement; (c) amplification of the reference sequence by polymerase chain reaction, using a P2 primer, a P4 primer with a 3 '' region capable of hybridizing to the reference sequence and a T region in the 5 'tail 'which is not complementary to the reference sequence, and a P5 primer that is capable of hybridizing to the reference sequence substantially adjacent, in the 3' 'direction, to the sequence capable of hybridizing to said 3' 'fraction of primer P4. |
