A human IL-2 mutein numbered according to wild-type IL-2 that has a substitution of an amino acid at one of positions 20, 88, and 126, wherein position 20 is substituted by an amino acid selected from the group consisting of: alanine, histidine, isoleucine, methionine, glutamine, serine, valine, or tyrosine position 88 is substituted by an amino acid selected from the group consisting of: alanine, arginine, glutamic acid, histidine, lysine, leucine, phenylalanine , glycine, isoleucine, methionine, serine, threonine, tyrosine or valine; position 126 is substituted by an amino acid selected from the group consisting of: asparagine, leucine, proline, phenylalanine, glycine, isoleucine, methionine, arginine, serine, threonine, tyrosine or valine; and whereby said IL-2 mutein preferentially activates T cells against Natural Killer cells. |
