PROCEDURE FOR PREPARING A HUMAN PAPILOMAVIRUS VACCINE WITH DISASSEMBLED AND REASSEMBLED VIRUS ANALOG PARTICLES. |
abstract |
A process for making a human papillomavirus vaccine comprising particles analogous to the HPV virus (VLPs), the method comprising the steps of: (a) incubating VLPs in a dissociation mixture comprising 2-20 mM dithiothreitol (DTT), a salt present in a NaCl equivalent range of 0.10M to 0.2M, a non-ionic surfactant, a metal chelating agent, and a buffer until disassembly of the VLPs occurs; (b) remove the reducing agent from the dissociation mixture; and (c) reassemble the disassembled VLPs. |
