THE PMK18 VECTOR IS CHARACTERIZED BY HAVING TWO FUNCTIONAL ORIGINS OF REPLICATION FROM ESCHERICHIA COLI AND THERMUS THERMOPHILUS, A GENE THAT CONFERS THERMOSTABLE RESISTANCE TO KANAMYCIN PROTECTED BY PATENT OF INVENTION 9301522 APPLIED FOR IN SPAIN, AN INDUCIBLE PROMOTER IN ESCHERICHIA COLI AND A CLONING REGION TWO MULTIPLE RESTRICTION TARGETS. WHEN THIS CONSTRUCTION IS INTRODUCED IN THERMUS THERMOPHILUS HB27 OR IN ESCHERICHIA COLI, A SOLUBLE ENZYME OF 192 KILODALTONS OF MOLECULAR SIZE IS SYNTHESIZED, CAPABLE OF CATALYSING THE REDUCTION OF NITRATE OR NITRITE AT AN OPTIMUM TEMPERATURE OF 80 C.
