USING GENETIC ENGINEERING TECHNIQUES, THE CARBAMATE KINASE OF ENTEROCOCCUS<PIX> (FORMERLY STREPTOCOCCUS<PIX>) FAECALIS<PIX> IS PREPARED, WHICH CATALYSES IN THE PRESENCE OF MG2+ THE FORMATION OF CARBAMATE AND ADENOSINE TRIPHOSPHATE (ATP), FROM CARBAMYL PHOSPHATE AND ADENOSINE DIPHOSPHATE (ADP), AS WELL AS THE REVERSE REACTION, FROM CARBAMATE AND ATP TO CARBAMYL PHOSPHATE AND ADP. A GENE CODING FOR CARBAMATE KINASE IS ISOLATED FROM THE USUAL SOURCE OF THE ENZYME ( ENTEROCOCCUS FAECALIS<PIX>), SEQUENCED, AND USED TO TRANSFORM A MICROORGANISM AND GENERATE A TRANSFORMANT THAT CAN BE CULTIVATED TO PRODUCE KINASE WITH HIGH YIELD. A PURIFICATION PROCEDURE IS USED THAT ALLOWS THE GENERATION OF ESSENTIALLY PURE ENZYME. |
