A PROCEDURE FOR THE OBTAINING OF ACETYLENE AND ALENIC DERIVATIVES OF 2-AMINOMETIL-5-METOXIINDOLES, OF PHARMACOLOGICAL INTEREST AS MONOAMINOOXIDASE INHIBITORS, IS DESCRIBED. THE PROCEDURE IS DEVELOPED IN THREE SUCCESSIVE STEPS: A) PART OF THE 1-H ACIDS (ALKYL OR ARYL) -5-METOXYINDOL-2-CARBOXYLICS THAT TREATED WITH ACID HALIDES LEAD TO THE CORRESPONDING ACID HALIDES, WHICH AMINES TO GIVE THE RESPECTIVE AMIDES (2); B) AMIDES ARE REDUCED WITH LITHIUM AND ALUMINUM HYDRORIDE TO THE RESPECTIVE AMINES (3), WHICH ARE ISOLATED AND CHARACTERIZED AS FREE BASES OR SOME OF THEIR SALTS; C) THE AMINES (3) OBTAINED ARE SUBMITTED TO A N-ALKYLL REACTION WITH ACETYLENE OR ALENIC HALIDES, IN THE PRESENCE OF TERCBUTILAMINE TO GIVE THE DESIRED ACETYLENE AMENES (4) WHICH ARE DESIRED AND CHARACTERIZED BY THE METHODS ORGANIC CHEMISTRY, EITHER AS SUCH OR IN THE FORM OF SOME OF ITS SALTS. THE AMINES (3) AND (4) HAVE BEEN STUDIED AS INHIBITORS OF MONOAMINOOXIDES A AND B AND DATA ARE PROVIDED ON THEIR ACTIVITIES AND SELECTIVITIES AS INHIBITORS OF THESE ENZYMES. |
