abstract | The invention relates to a method for detection and/or quantification of at least one nucleic acid sequence in a sample, comprising the steps of: (a) optionally measuring the nucleic acid concentration in the sample; (b) amplifying the at least one nucleic acid sequence by nested polymerase chain reaction using nucleic acids comprised in the sample as template to form a set of first amplicons comprised in a first reaction mixture; (c) optionally measuring the nucleic acid concentration in the first reaction mixture; (d) amplifying the at least one nucleic acid sequence by polymerase chain reaction using the set of first amplicons as template and using nucleotide primers which are specific for the at least one nucleic acid sequence and which are coupled to magnetic beads to form a set of bead-coupled second amplicons comprised in a second reaction mixture; (e) optionally isolating the magnetic beads from the second reaction mixture; (f) combining the magnetic beads from step (d) or (e) with a labeled probe, wherein the probe binds specifically to the nucleic acid sequence; (g) detecting and/or quantifying magnetic beads labeled with the probe; and (h) determining the presence and/or the amount of the at least one nucleic acid sequence in the sample based on the detection and/or quantification in step (g). Further, the invention relates to a device and system for carrying out this method. |
