abstract | The present invention relates to a method of sequencing a nucleic acid molecule, the method comprising in particular: (a) providing the immobilized nucleic acid on a surface; (b) forming a single-strand breccia section by partial duplexing of the nucleic acid such that the single-strand breccia section is flanked by duplex sections, the sequence to be sequenced being a sequence of the breccia section single strand; (c) providing a set of at least four fluorescently labeled oligonucleotide probes; (d) detecting the binding, or lack thereof, of the fluorescently labeled oligonucleotide probes with the gap section of the immobilized nucleic acid, the identity of the interrogated nucleotide of the gap section of the immobilized nucleic acid being identified as the complementary base of nucleotide X of the fluorescently labeled oligonucleotide probe which has the highest incidence of attachment; (e) repeating steps (c) and (d) to interrogate subsequent nucleotide positions of the gap section of the immobilized nucleic acid until sufficient nucleotides of the gap section have been identified to determine a sequence. The invention also relates to methods of phenotyping and monomolecular sequencing; phenotyping and monomolecular identification methods of a nucleic acid labeled molecule; an immobilized nucleic acid molecule; and a composition comprising oligonucleotide probes. |
