abstract The invention relates to a method for detecting one or more polynucleotide amplification products, the method comprising, in one embodiment, the steps of: a) providing one or more oligonucleotide primer groups, each group comprising i) a forward primer comprising a fluorescence cassette specific portion and a downstream target specific portion 3 "; and ii) one or more target specific primers; b) providing one or more fluorescent pairs extinguished, each wherein said pair is characterized by: i) a first cassette oligonucleotide labeled with a fluorescent moiety and having a sequence that is capable of hybridization with the complement of the fluorescence cassette specific portion of the primer before a group of oligonucleotide primers, and ii) a second cassette oligonucleotide labeled with a quencher moiety; c) initiating an amplification reaction; isothermal polynucleotide in the presence of a strand displacement DNA polymerase, thereby generating a sequence complementary to at least a portion of the relevant forward primer, such that the second relevant cassette oligonucleotide is less able to hybridize to the first relevant cassette oligonucleotide, whereby a signal is generated; and d) detecting the signal.
