abstract In vivo methods, compositions and kits for producing nucleic acids that include at least one unnatural base are provided. The invention provides in vivo methods for producing a nucleic acid that contains an expanded genetic alphabet, the method comprising incorporating at least one non-naturally occurring base into the nucleic acid. The invention relates to semi-synthetic organisms comprising an expanded genetic alphabet, the genetic alphabet comprising at least one non-natural base. The present invention provides compositions comprising a heterologous or recombinant polymerase and methods of using the same. In addition, the invention provides kits that are useful for stably incorporating non-naturally occurring nucleic acid into a nucleic acid molecule, for example, using the methods provided by the present invention in an in vitro state or in a cell-free state.
