abstract | 1. The application describes a method of calibrating a sample heating device, comprising: nproviding a sample that includes a temperature-sensitive reagent that emits a luminescent signal in response to a stimulus; nwherein the luminescent signal emitted from the temperature-sensitive reagent changes as a function of temperature in a known and predictable manner and is not directly proportional to an amount of nucleic acid present in the sample, nstimulating the temperature-sensitive reagent to induce emission of the luminescent signal therefrom; ndetermining a luminescence-determined temperature of the sample based on the luminescent signal emitted by the temperature-sensitive reagent; ndetermining a device-determined temperature for the sample; and nadjusting a temperature setting of the sample heating device based on at least one of the luminescence-determined temperature and device-determined temperature. |
