abstract | The invention relates to compositions and methods for the manufacture and optimization of modified mRNA molecules via optimization of their terminal architecture. Described herein are compositions and methods for the manufacture and optimization of modified mRNA molecules via alteration of the terminal architecture of the molecules. Specifically disclosed are methods for increasing or altering protein production or localization by altering the 5 'UTR of modified mRNAs. In one aspect, provided is a synthetic isolated RNA comprising a first region of linked nucleosides encoding a polypeptide of interest, a first flanking region located at the 5' terminus of the first region, a second flanking region located at the 3' terminus of the first region and a 3' tailing region of linked nucleosides. Any of the first region, first flanking region, second flanking region or a 3' tailing region may comprise at least one modified nucleoside. |
