abstract | The present invention relates to an expression cassette (also referred to herein sometimes as "expression system" or "system") comprising (a) a first nucleotide sequence encoding a polypeptide which is secreted from a fungal host cell; (b) a second nucleotide sequence encoding a polypeptide of interest; and (c) one or more third nucleotide sequences which can be bound by a polypeptide comprising at least one sequence specific RNA binding domain,wherein the order (5' → 3') of said first (a), second (b) and third (c) nucleotide sequence(s) is as follows: (i) (a), (b) and (c); (ii) (a), (c) and (b); (iii) (b), (a) and (c); (iv) (b), (c) and (a); (v) (c), (a) and (b), or (vi) (c), (b) and (a), wherein the nucleotide sequence of (a) and (b) or (b) and (a) are fused in frame. Also contemplated are vectors comprising the expression cassette, host cells comprising the vectors as well as methods and uses for the production of a polypeptide of interest. |
