abstract | The present invention pertains to a method for synthesizing a combinatorial library comprising the following steps: (i) providing a library of template nucleic acids that are partially double-stranded, wherein each template nucleic acid comprises a reactive group, and wherein the two strands in the double-stranded region are covalently linked, (ii) contacting said template nucleic acids with a library of adapter oligonucleotides, wherein each adapter oligonucleotide of the library comprises a first sequence of 2 or more defined nucleotide residues at one end, and a second sequence of from 7 to 60, nucleotide residues, (iii) ligating those of the adapter oligonucleotides to the last nucleotide of the double-stranded portion of the template nucleic acid which have hybridized with their first sequence to the single stranded portion of the template nucleic acid in immediate proximity to the double-stranded portion of the template nucleic acid, (iv) sorting said template nucleic acids according to the second sequences of the adapter oligonucleotides, (v) chemically modifying said reactive group of the template nucleic acids, (vi) removing the second sequence of the adapter oligonucleotide by cleavage of the ligated adapter oligonucleotides between the first sequence and the second sequence or by enzymatic digestion with an exonuclease, and (vii) repeating steps (ii) to (vi) as required. |
