titleOfInvention High throughput analysis of T-cell receptor repertoires
abstract A method for analysis of a patient's T cell repertoire is provided, whereby nucleic acid molecules encoding T cell receptor protein sequences are obtained from lymphocytes isolated from a patient, a plurality of said nucleic acid molecules obtained is sequenced, rendering sample TCR sequences, the sample TCR sequences are aligned with a TCR sequence data base, and sample TCR sequences positively aligning to data base sequences are selected as selected sample TCR sequences, and the frequency distribution of at least some of the selected sample TCR sequences is determined. A method is preferred whereby the TCR sequence data base is obtained by stimulating lymphocytes isolated from a patient in vitro with one or several antigens, selecting T cells responding to stimulation from said lymphocytes and obtaining a plurality of nucleic acid sequences encoding T cell receptor proteins from at least some of the T cells selected.
