abstract | The object is to provide a method for confirming the amplification of a nucleic acid rapidly, in a simple manner, at low cost and at high sensitivity after an amplification reaction of the nucleic acid. Specifically, disclosed is a DNA fragment which is a single-stranded DNA fragment having a hairpin structure formed therein, wherein a bulge structure is formed in the hairpin structure. The DNA fragment is intended to be used in the form attached to the 5'-terminus of a nucleic acid amplification primer. Also disclosed is a method for confirming the amplification of a nucleic acid, which comprises conducting a nucleic acid amplification reaction such as PCR by using a hairpin primer having the DNA fragment attached to the 5'-terminus thereof, and measuring the amount of the hairpin primer by using a fluorescent molecule having a bulge structure attached thereto. For example, the method for confirming the amplification of a nucleic acid can be applied to an allele-specific PCR to detect an SNP rapidly, in a simple manner, at low cost and at high sensitivity. |
