abstract | The present invention describes an improved method of generation of Brassica transformants based on a method of selecting genetically transformed Brassica juncea explants based on their ability to utilize Xylose as a sole carbohydrate source. The said invention encompasses the process of Agrobacterium Mediated Transformation of the target host plant with the constructed vector for the expression of the enzyme Xylose Isomerase. Also disclosed is the method of selecting the putative transformants subsequent to transformation with the said vector to obtain a metabolic advantage of utilizing Xylose as a carbon source. The subject invention alleviates the disadvantage of the negative selection methods. Reported is a stable transformation efficiency of 34-40% using Xylose Isomerase for selection. |
